Strains were grown in 50 mL of YPD until exponential phase (OD=0.8) and then 1 mM sodium selenite was added for 30 minutes from a 0.2 M stock solution (diluted in water). Strains were grown in 50 mL of YPD until exponential phase (OD=0.8) and then 2 mM cadmium chloride was added for 30 minutes from a 0.2 M stock solution (diluted in water). Strains were grown in 50 mL of YPD until exponential phase (OD=0.8) and then centrifuged, washed once in sterile water and resuspended in YPD containing 1 M NaCl at 30°C for 30 minutes. Strains were grown in 50 mL of YPD until exponential phase (OD=0.8) and then 5 mM iron sulfate was added for 30 minutes from a 0.5 M stock solution (diluted in water). Strains were grown in 50 mL of YPD until exponential phase (OD=0.8) and then 0.5 mM bathophenanthroline disulfonate was added for 30 minutes from a 0.2 M stock solution (diluted in water). Strains were grown in 50 mL of YPD until exponential phase (OD=0.8). Total RNA was extracted using the RNA easy kit (Qiagen) and following the supplier's instructions. One microgram of RNAs from the two samples to be compared were reverse-transcribed for each independent preparation and labeled with Cy3 or Cy5 dye using the indirect labeling procedure described on www.transcriptome.ens.fr. The pooled Cy3 and Cy5 labelled cDNA were hybridized using the Agilent recommendations. The arrays were scanned using an Agilent scanner with a 2 microns resolution. The features were extracted using the feature extraction software with a standard Gene Expression protocol.
